Abstract:

Objective: The Hippo pathway plays an important role in embryo development, and separation of trophectoderm (TE) and inner cell mass (ICM) cell lines. Therefore, this study investigated effect of maternal age on activity of Hippo pathway in human embryos. Materials and Methods: In this experimental study, the developed up embryos to the blastocyst stage and the embryos whose growth stopped at the morula stage were collected from women aged 20-30 years old (young group, 94 embryos) and >37 years (old group, 89 embryos). Expression of OCT4, SOX2, CDX2, GATA3, YAP genes and the relevant proteins, in the both groups were evaluated using respectively quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and immunofluorescence methods. Results: There was no significant difference in the expression level of OCT4, SOX2, CDX2, GATA3 and YAP genes in blastocyst and morula stages, between the two groups. However, SOX2 and CDX2 gene expressions in morula stage embryos of the old group was statistically lower than that of the young group (P=0. 007 and P=0. 008, respectively). Additionally, in the embryos collected from women with >37 years of age, at the blastocyst stage, phospho-YAP (p-YAP) protein was found to be accumulated in the TE, but it was almost disappeared from the ICM. Additionally, in the old group, contrary to the expectation, YAP protein was expressed in the ICM, rather than TE. Conclusion: The results of this study showed that YAP and P-YAP among the Hippo signalling pathway may be altered by increasing age.

Abstract:

The husseiniye is a religious place in Shia Islam, where the mourning ceremony of Imam Hussein (PBUH) and the martyrs of the Battle of Karbala is mainly held. Apparently, the husseiniye has come into existence as an extension of the structures of the mosque and religious places such as the zaviye and the tekiye and as a result of integration and reproduction of parts of their structures and functions. Many concepts have been lost in the design of the contemporary husseiniye as a single building, which is open to the public only in the months of Muharram and Safar. The husseiniye and the tekiye are among the entities that are responsible for the important celebration of the Battle of Karbala. Sanctuaries have always had a particular status as internal organs of any city or village, and religious buildings have always been regarded as one of the most fundamental components of the fabric, structure, and shape of a city. Therefore, the mosques, religious buildings, worship centers, and social and cultural centers in a city constitute a long-standing identity that can never be separated from its social and urban structure. Moreover, man is a social being, and needs social interactions and communication with others. Urban spaces play a significant role in the realization of this characteristic, and the courtyards of mosques and religious places such as the husseiniye are among these spaces. The Iranian husseiniye or tekiye is a symbol of the link between time, place, and people, with an identity distinct from that of public places with the same function in other countries due to its quality as an urban space. It is also unique as the only open urban space with a religious function in the Islamic urban development culture. Given the cultural characteristics of our country, particularly the role of Islam and religious beliefs in Iranians’ national and social identity, the necessity of investigating the physical qualities of the husseiniye seems obvious. The purpose of this research was to recognize the effect of the Ashura rituals on the configuration of the city of Na'in, Iran based on the pathway pattern. On that basis, it can be stated as the research hypothesis that the pathway pattern of the groups of Ashura mourning, routinely held in the city, has such a significant effect on the formation and spatial organization of the city and locations of the husseiniye buildings that it cannot be disregarded in their analysis. According to what was stated, the research question concerns the aspects of the social and religious identification by the rituals in Na'in. Therefore, we conducted a structured study of seven husseiniye buildings in Na'in to examine the physical indicators of the husseiniye and their adaptation to the factors influencing the formation of buildings as open urban spaces and to analyze human pathay in the husseiniye using the pathway pattern. The research results indicate that the factors effective on consideration of open religious places like the husseiniye as open urban spaces include the general quality of convenience and communication with the surrounding passages, multiple entrances, the interaction established between the people, and urban life spirit. Thus, Nowgabad Mosque has the greatest number of alternative pathway patterns with six entrances, and the Husseiniye of Gowdalu has the lowest with two entrances. Better recognition and employment of the principles for design of such spaces, including location, flexibility in form and function, and rich symbolic concepts, can greatly contribute to persistence of the physical expression of a culture in the environment and identification of the Iranian city.

Abstract:

Background: Among the primary bone tumors, osteosarcoma accounts for a malignant tumor with a high rate of progression and poor prognosis. Despite the achievement of combined therapy regimens in improving patients’,overall survival, patients with osteosarcoma confront the chemoresistance obstacle. Objectives: This study aimed at determining the expression pattern of autophagy and necroptosis pathways mediators in osteosarcoma tumors. Methods: The expression level of autophagy main mediators such as autophagy-associated protein 5 (ATG5), Beclin 1 (BECN1), and microtubule-associated protein 1A/1B-light chain 3 (LC3), necroptosis biomarkers such as receptor-interacting protein kinases (RIPK1 and RIPK3), and mixed lineage kinase domain-like (MLKL) were evaluated in 80 bone tissues including 60 bone tumors (40 malignant tumors and 20 benign tumors) and 20 margin tissues, using real-time PCR. The correlations of gene expression levels with the patient’, s clinical and pathological features were considered. Results: Based on our data, ATG5, BECN1 and LC3 expression were down-regulated in osteosarcoma tumors compared to margin tissues. Also, malignant osteosarcoma tumors showed a significant decrease in the expression level of RIPK1 andMLKLas necroptosis regulators, which revealed a correlation with tumor malignancy. In addition, the higher expression levels of BECN1, LC3, RIPK1, and MLKL were observed in tumor tissues of patients under the chemotherapy regimen, indicating the relevance of autophagy and necroptosis pathways with the patient’, s response to therapy. Conclusions: Reduction in the expression level of autophagy and necroptosis mediators in high-grade osteosarcoma tumors indicates the possible impact of these pathways on the rate of proliferation and growth of osteosarcoma tumor cells and can emphasize the importance of cell death alternative pathways for treatment when apoptosis machinery is mutated and cause chemoresistance.

Abstract:

The presence of water resources, particularly rivers, significantly influences site selection. Throughout history, settling near rivers has brought numerous advantages, but it has also posed certain risks. This study focuses on the sub-basins southeast of the Caspian Sea. Initially, we explore the connection between the layout of ancient settlements and the drainage networks. Additionally, we analyze the relationship between flood risk and the settlement patterns of these ancient sites by estimating the flooding risk based on linear, areal, and relief aspects. This research marks the first time such an analysis has been conducted. The findings underscore the importance of proximity to rivers in site selection, with areas close to rivers with lower stream orders being the most favorable for settlements. The study reveals a decrease in the frequency of sites near higher stream orders in relation to flooding risk. Conversely, there is an increase in the frequency and density of ancient sites near the first stream orders and at greater distances from the rivers, coinciding with an elevated flooding risk in the sub-basins. These results indicate that the inhabitants of the southeastern areas of the Caspian Sea sub-basins were cognizant of the flooding danger and factored it into their decision-making when selecting settlement sites

Abstract:

Purpose: The aim of the present study was to investigate possible side effects of botulinum toxin on the visual pathway, measured by visual evoked potentials using two stimulation techniques: flash and pattern reversal checkerboard. Patients and Methods: Twenty-six eyes from thirteen female subjects, aged between 40 and 52 years, who received botulinum toxin injections for cosmetic purposes in the upper face area, comprised the case group. Visual evoked potentials using two stimulation techniques (flash and pattern reversal checkerboard) were measured in this group. The obtained results were compared with those of 26 eyes from 13 healthy females with no history of botulinum toxin, who formed the control group. Results: A significant difference was observed in the latency of the flash visual evoked potential P100 peak between the case and control groups (P<0. 001) when using the flash stimulation technique. Conclusion: Botulinum toxin may affect the visual pathway of patients, which can be measured using flash visual evoked potentials.

Abstract:

Black cumin (Nigella sativa) is an important medicinal plant belonging to the Ranunculaceae which produces large diversity of secondary metabolites especially terpenes. Mono (C10) and tri-terpenes (C30) are two major structural classes of terpene compounds. Several genes are involved in biosynthetic pathway of the monoterpenes known as MEP pathway, located in plastids. In the MEP pathway, monoterpene synthases are rate- limiting enzymes for monoterpene biosynthesis. In the biosynthetic pathway of monoterpenes geranyl diphosphate synthase catalyzing geranyl diphosphate (GDP) biosynthesis as universal precursor of monoterpenes. Oxidosqualenecyclases is of the most important. In this study gene expression patterns of a monoterpene synthase, GDP synthase, beta amirin synthase and squalene oxidase were evaluated under salicylic acid treatment. Gene expression analysis of the genes at trasnscript level using semi-quantitative PCR in response to salicylic acid treatment revealed their differential expression.

Abstract:

Objective: Ajowan (Trachyspermum ammi L. ) is an annual plant in Apiaceae family which is used in traditional medicine and medical science. This study was carried out to determine the quantitative expression pattern of some genes on monoterpenoid biosynthesis pathway and essential quality in Ajowan treated by methyl jasmonate. Ajowan essential oil compounds and the amount of MECPS, DXR, and GDs genes expression as main genes related to Thymol biosynthesis pathway in response to methyl jasmonate in 4 different times were analyzed. Materials and methods: After flowering, the 0. 1 mM methyl jasmonate hormone was sprayed onto the plants. Samples were taken in 4 times (after 8, 24, and 48 treatment and control treatment). RNA extraction, cDNA synthesis, and Real-Time PCR have done from flowers and the date from these gene expressions were analyzed. Essential chemical compounds were studied by GCGC/ MS. Generally, in 4 samples (samples were taken after 8, 24, and 48 treatment and control treatment), 10 different compounds were distinguished. Results: Based on results, the study of the essential oils of Ajowan showed that the changes of the amount of secondary compounds in the essential oil, the amount of γ-Terpinene (precursor of monoterpenoids like Thymol) went up after 24 hours methyl jasmonate treatment. Due to Real-Time PCR dates, the quantity of all 3 studied genes increased after 24 hours methyl jasmonate treatment. The increasing of these genes expression can be causes the incline level of γ-Terpinene in essential oil. Conclusions: Increasing the amount of 3 mentioned genes expression and also a high amount of γ-Terpinene in essential oils after 24 hours in flowers show that this hormone can affect the gene expression in MEP pathway which causes monoterpenoids production increase after one day of using the hormone.
